TASKS. The contractor shall:

• Design and optimize recombinant protein expression vectors for various host systems e.g., Mammalian/Expi293/CHO, Insect/Baculo
  

• Perform in silico design of chimeric proteins, including the strategic selection of fusion tags HIS, Fc , protease cleavage sites TEV, HRV-3C , and signal peptides 
  

• Perform in vitro biochemical and biophysical assays to evaluate proteins and constructs (e.g., SDS-PAGE, Western Blotting, UV-Vis
  (NanoDrop), SEC-HPLC) 
  

• Perform and evaluate recombinant protein-antibody interactions and identify biologically relevant findings (e.g., ELISA, BLI)
  

• Operate and maintain FPLC systems (e.g., Cytiva ÄKTA Pure/Start) and associated software (UNICORN).
  

• Operate high-throughput Biolayer Interferometry (BLI) and Surface Plasmon Resonance (SPR) instruments (e.g., Octet RH96, Carterra, LSA)
  

• Utilize specialized analytical software (e.g., Octet Analysis Studio, Carterra Analysis Software, GraphPad Prism) for advanced kinetic
  modeling and epitope binning determination
  

• Conduct structure prediction and modeling as needed to inform experimental design (AlphaFold)

REQUIREMENTS. The contractor must have:

• Master’s in Biology, life sciences or related field
  

• 7-8 years experience in biology or related field performing biological research functions in a research setting and laboratory environment.
  

• Demonstrate knowledge of proper laboratory disposal methods and safety procedures.
  

• Experience maintaining and utilize cell lines for protein expression, and perform downstream supernatant harvest, cell lysis, and clarification
  

• Expertise developing and optimize multi-step protein purification protocols to improve yield, purity, and stability using diverse purification and chromatography techniques
